چکیده

Multiple mechanisms have been proposed to account for immune escape by tumors. Although gangliosides have long been known to suppress T-cell immunity, few studies have examined the effect of human tumor-derived gangliosides on immune responses. Here, we show that gangliosides isolated from renal cell carcinoma (RCC) cell lines and clear cell tumor tissue can induce apoptosis in peripheral blood T cells. The RCC tissue-derived gangliosides also suppressed IFN-; and, in many cases, interleukin-4 production by CD4 T cells at concentrations (1 ng/mL-100 pg/mL) well below those that induce any detectable T-cell death (4-20 Mg/mL). Additional findings show that GM2 expressed by RCC plays a significant role in promoting T-cell dysfunction. This is supported by the demonstration that all RCC cell lines examined (n = 5) expressed GM2 as did the majority of tumors (15 of 18) derived from patients with clear cell RCC. Furthermore, an antibody specific for GM2 (DMF10.167.4) partially blocked (50-60%) T-cell apoptosis induced by coculturing lymphocytes with RCC cell lines or with RCC tissue-derived gangliosides. DMF10. 167.4 also partially blocked the suppression of IFN-; production induced by RCC tissue-derived gangliosides, suggesting that GM2 plays a role in down-regulating cytokine production by CD4 T cells. Downregulation of TMEM40 by miR-138-5p suppresses cell proliferation and mobility in clear cell renal cell carcinoma

Background: Clear cell renal cell carcinoma (ccRCC) represents approximately 70% of RCC,as the most frequent histological subtype of RCC. MiR-138-5p, a tumor-related microRNA (miRNA), has been reported to be implicated in the diverse types of human malignancies, but its role in ccRCCremains unclear. Objective: The study was designed to investigate the function...

Comparison of COX2 expression in radiation induced basal cell carcinoma and non-radiation induced basal cell carcinoma

Background: Radiation-induced basal cell carcinoma (BCC) can be multiple, large, and recurring, which complicates its treatment in some cases. According to reports on the role of cyclooxygenase 2 (COX2) inhibitors in the treatment or prevention of non-melanoma skin cancers and considering the fact that COX2 expression has not been evaluated in radiation-in...

Prognostic value of p53 in renal cell carcinoma

Background and Objectives: RCC is one of the most common genitourinary cancers. Accurate prediction of prognosis would be valuable for adjuvant trial design, counseling and effectively scheduling follow up visits. P53 is a tumor suppressor gene that expresses a protein that involved in both cell-cycle ar...

Expression Of P53 And Ki67 Proteins In Renal Cell Carcinoma And Its Relationship With Nuclear Grade

Background and Objective: Evaluation of tumor proliferative activity may provide a predicting parameter to estimate biologic aggression and a subsequent prognosis that has been evaluated in many malignancies. We have selected renal cell carcinoma (RCC) in this study. To determine tumor proliferative activity, KI67 antibody was applied and results were compared with apoptosis, applying P53 antib...
